چکیده
Blautia has attracted attention because of its potential efficacy in ameliorating host energy metabolism and inflammation. This study aims to investigate the influences producta D4 on colitis induced by dextran sulfate sodium (DSS) reveal underlying mechanisms. Results showed that B. producta intervention significantly relieved body weight loss, suppressed elevation pro-inflammatory cytokines [including interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β)] excessive oxidative stress [myeloperoxidease (MPO) activity, superoxide dismutase (SOD) glutathione peroxidase (GSH-Px) malondialdehyde (MDA) level] mice. Moreover, concentrations tight junction proteins (occludin, claudin-1, ZO-1) related intestinal barrier were obviously elevated, colitis-related TLR4/NF-κB pathway activation was remarkably inhibited after intervention. The microbial disorder evidently ameliorated increasing relative abundance Clostridium sensu stricto 1, Bifidobacterium, GCA-900066225, Enterorhabdus, reducing Lachnospiraceae NK4A136 group. In conclusion, oral administration could ameliorate DSS-induced suppressing inflammatory responses, maintaining barrier, inhibiting TLR4/ NF-κB pathway, regulating microbiota balance. These results are conducive accelerate development as a functional probiotic for colitis.

Calycosin attenuates dextran sulfate sodium (DSS)-induced experimental colitis
Objective(s):Inflammatory bowel disease (IBD) results from dysregulation of intestinal mucosal immunity. It is an incurable disease that affects millions of people worldwide. Developing new strategies for the treatment of colitis has been a major challenge. Here, we report the effect of calycosin, a plant-derived flavonoid, in successfully managing colitis in murine model. Material and Methods:...
متن کاملAlloferon Alleviates Dextran Sulfate Sodium-induced Colitis
Dysfunction of gut immune regulation is involved in mucosal damage in inflammatory bowel disease (IBD). However, there is still no efficacious immune-regulator for the treatment of IBD. Alloferon is a novel immune-modulatory peptide that was originally isolated from infected insects. It shows anti-inflammatory effects by the regulation of cytokine production by immune cells and their activities...
متن کاملProtein Kinase D2 Protects against Acute Colitis Induced by Dextran Sulfate Sodium in Mice
Inflammatory bowel disease is characterized by dysregulation of the mucosal immune system resulting from impaired intestinal epithelial barrier function. Protein kinase D2 has been implicated in the regulation of immune responses. The present study was to define PKD2 might affect murine colitis. Colitis was induced in wild-type mice (PKD2WT/WT) and PKD2 catalytic activity deficient mice (PKD2SS...
متن کاملBortezomib protects against dextran sulfate sodium-induced ulcerative colitis in mice
Bortezomib, a first‑in‑class proteasome inhibitor, is a standard method of treatment in multiple myeloma. In the present study, the therapeutic effect of bortezomib was evaluated in an ulcerative colitis model induced by dextran sulfate sodium (DSS) in mice, and the mechanism of action was also investigated. Mice were administered with 3% DSS drinking water for 7 consecutive days and then they ...
متن کاملProbiotic administration alters the gut flora and attenuates colitis in mice administered dextran sodium sulfate.
BACKGROUND Probiotics are used in the therapy of inflammatory bowel disease. This study aimed to determine whether prior administration of probiotic lactobacilli and bifidobacteria would prevent disease and change gut flora in an animal model of colitis. METHODS Swiss albino mice received a probiotic mixture (four Lactobacillus and four Bifidobacterium species) or medium (control) for a week ...
